The main goal of the cancer research is to improve the clinical outcome of cancer patients.At present,we can perform tailor-made chemotherapy,the selection of effective chemotherapy based on evaluation of tumor-associated biomarkers.For example,5-fluorouracil (5-FU)-derived agents such as S-1 against tumors with a low expression of thymidylate synthase (TS),gemcitabine against tumors with a low expression of ribonucleotide reductase subunit M1 (RRM1),cisplatin against tumors with a low expression of ERCC 1,and tyrosine kinase inhibitors of EGFR against tumors with EGFR gene mutations.However,the remaining populations of cancers,with high expressions ofTS,RRM1,ERCC1,and a wild-type of EGFR,need other new treatments.Among them,TS plays a central role in the biosynthesis of thymidylate,an essential step in DNA synthesis,and its expression is associated with the cell proliferation.
