The construction of one longer genetic linkage map based on double haploid population of broccoli

  Broccoli is a major crop in Brassica oleracea and with higher quality,medical value,especially being widely studied in the world.Two advanced inbred lines of broccoli,86101 and 90196,were used as the parents to develop a double haploid(DH)mapping population by microspore culture,the population contained 176 lines plant.314 pairs of SSR primers were selected from 3786 pairs of EST-SSR and g-SSR primers,and all the primers were proved with good polymorphism,stable,repeatable.A longer genetic linkage map was constructed by the software of Joinmap 4.0,the linkage map included 268 SSR markers and spanned 1293.32 cM with an average of 4.8 cM in nine linkage groups.In the linkage map,the genetic distance of nine linkage groups ranged from 26.9 cM to 302.8 cM,the longest linkage group was LG1 spanning 302.8 cM with an average of 5.1 cM,and the shortest linkage group was LG7 spanning 26.9 cM with an average of 1.7 cM,the two linkage groups contained respectively 60 and 16 pairs of SSR primers.This study will help to molecular assisted selection in broccoli.
