Evi5 is Required for Multi-System Regeneration in Xenopus and Axolotl

  Amphibians such as the Salamanders and the African clawed frog Xenopus are good models for regeneration studies because they can fully regenerate their lost organs.While axolotl can regenerate damaged organs throughout its life, Xenopus has limited regeneration capacity after metamorphosis.The ecotropic viral integrative factor 5 (Evi5), a cell-cycle-regulated protein that can prevent cells from entering the mitosis prematurely, is of great interest for it is highly upregulated in axolotl limb regeneration blastema, but remains unchanged in the fibroblastema of postmetamorphic frogs.Here we show that Evi5 is also up regulated after limb and the tail amputations in Xenopus tadpole, as in axolotls.Down regulation of Evi5 with morpholino antisense oligos impairs wound healing and blastema formation in the limb and tail in both axolotl and Xenopus tadpoles, suggesting that Evi5 has conserved functions in organ regeneration.In addition to its involvement in cell cycling, we are currently using skin punch wounding as a healing model to investigate whether Evi5 is involved in cell migration during regeneration.Our results suggest that Evi5 plays critical roles in regeneration of multi-systems in amphibians.
