Background: Diabetic wounds is a major clinical challenge, because minor skin wounds can lead to chronic, nonhealing ulcers and ultimately result in infection, gangrene, or even amputation.Studies on bone marrow-derived mesenchymal stem cells (BMSCs) and a series of growth factors show many benefits to the wound healing and regeneration.And platelet-rich plasma (PRP) secretes at least twenty growth factors may represent a better environment for BMSCs development and differentiation.Whether combined use of BMSCs and PRP may be more effective in accelerating diabetic ulcer healing remains unclear.Objective: The aim of present study is to investigate the efficacy of BMSCs associated with PRP in the repair of refractory wound healing in diabetic rats model.Methods: Forty-eight male Sprague-Dawley rats with diabetes mellitus induced by streptozotocin (DMIS rats) were divided into 4 groups: BMSCs plus PRP;BMSCs alone;PRP alone;phosphate buffered saline (PBS) as control.The rate of wound closure was quantified.Histopathological study was obtained regarding the wound depth and edge of the skin on 7, 14 and 28 days after surgery.Results: Wound healing rates were significantly higher in BMSCs plus PRP-treated group than other treatment groups.The immunohistochemistry results showed that PECAM-1, PCNA and TGF-β1 expression were significantly increased in BMSCs plus PRP-treated group as compared with other treatment groups.On day 7, the CD68 expression significantly increased in BMSCs plus PRP-treated wounds, and it marked decrease at 14 day compared with control group.Conclusions: The combination of BMSCs and PRP had a favouring effect on diabetic wound repair and regeneration.
