Many proteins function in the form of large protein complexes.The N-CoR (nuclear receptor corepressor) and the related factor SMRT (silencing mediator of retinoid and thyroid hormone receptors) were initially identified as corepressors for nuclear hormone receptors (NRs).N-CoR and SMRT are now known to play important roles in development,differentiation and tumorigenesis and serve as corepressors for many other transcriptional factors in addition to nuclear receptors.Over last several years studies from our group and others have clearly established N-CoR and SMRT as the class I HDAC-containing corepressor complexes.The stable SMRT/N-CoR core complex contains HDAC3,TBL1 and TBLR1,and GPS2.
