目的 The purpose of this study is to demonstrate the inhibitory effect of arsenic trioxide(As2O3)on lung cancer xenograftsgrowth and angiogenesis,and to explore the underlying mechanism.方法 Non-small cell lung cancer(NSCLC)and small cell lung cancer(SCLC)xenograft tumors models were built with lung cancercells alone or combined with matrigel.Then animals were treated with 2.5 or 5.0 mg/kg of As2O3,control drug Sorafenib ornormal saline.Tumor volumes and angiogenesis condition were compared among different groups.Ultrastructures of vascularendothelial cells were observed by electron microscope.Expression of angiogenic factors such as VEGF-A,VEGFR-2,HIF-1α,Dll4and Notch-1 were detected.Angiogenic factors were also detected in lung cancer cell lines and human umbilical vein endothelialcells(HUVECs)in vitro.
