Dendrimers have been extensively used for cancer drug delivery owing to their unique highly branched but precise molecular structures,high densities of surface groups allowing for various functionalizations,internal cavities for host-guest encapsulations,unusually low intrinsic viscosity facilitating transport in blood,and fast cellular uptake for biodelivery.However,how to efficiently synthesize such macromolecules at large scales and how to maintain their precise structures when used as carriers are key to their clinical translation.Herein,we will report new approaches to efficient synthesis of well-defined dendrimers.Particularly,we were able to obtain 240 KDa dendrimers in four steps.These dendrimers have been used as nanomedicine carriers for drug delivery and MRI imaging.Furthermore,we will report the first dendrimer conjugates with precise structures having strong anticancer activities(DendriDrug)and the dendrimer showing inherent anticancer activity without drug conjugation(Therapolymer).
