核酸分子-细胞膜作用过程中磷脂双层膜的电荷反转与重构

  界面核酸分子的结构与动力学是决定核酸药物稳定性与功效的关键.本实验室研发了一套偏振分辨和频振动光谱(SFG-VS)系统用于生物界面的原位、实时、免标记的检测.利用该光谱检测系统,我们对核酸分子在层状正电荷DMTAP 磷脂双层膜上的分子构相与吸附动力学进行了研究.PO2-波段,OH/NH 波段和“指纹区”波段的光谱数据显示,核酸分子的吸附在375 nM(ρ ≈ 3.75)达到饱和,取向角也趋于稳定.
