Directional cell migration and tumor invasion requires the tight coordination of actin assembly and plasma membrane remodeling, which is orchestrated by growth factor signaling. However, the molecular mechanism underlying this coordination remains elusive. The exocyst is an evolutionarily conserved octameric protein complex that regulates secretory vesicle docking and fusion during exocytosis.Recent studies demonstrate a role of the exocyst in cell migration and tumor invasion. In particular,Exo70, a component of the exocyst complex, directly interacts with the Arp2/3 complex, and kinetically stimulates actin polymerization and branching at the plasma membrane. In addition,Exo70 physically induces membrane curvature and generates protrusions on the surface of cells.
