Analysis of signal transduction networks controlled by TNF,IL-1,LPS and in turn,controlling immediate-early gene expression patterns in response to injury and infection,continues to attract a high level of interest,particularly with recent findings that the TIR domain containing Toll proteins are a crucial component of the innate immune system.Broadly,such stimuli activate MAP/stress kinases and the related pathways leading to Rel/NF*B family transcription factors.A number of components,for example IRAK-1,IRAK-2,IRAK-m,TRAF-s,MyD88,TAK-1/TAB-1 and TPL-2 have been identified,by a range of techniques,as playing a role in regulating/activating these pathways.
