The role of the insulin-like growth factors (IGF) system in tumorigenesis and tumor metastasis has been debated for many years but recent data appear to validate it as a promising target for cancer treatment.Currently, at least seven antibodies targeting at receptor (IGF-IR) are under early clinical trials.Access to IGF-IR on surface of tumor cells is essential for such antibody to have efficacy.However, penetration of large IgG molecule into solid tumors is an inherent difficulty.In an attempt to identify an optimal strategy for inhibition of IGF-mediated signal transduction leading to enhanced cancer cell proliferation we have been developing a panel of novel human monoclonal antibodies (hmAbs) against components of the IGF system, with focus on ligands.We have hypothesized that signal blocking of IGF system can be achieved by the depletion of soluble ligands, which may not require penetration of therapeutics.
