Acetyl-coenzyme A carboxylase (ACC) plays a crucial role in fatty acid metabolism, and its inhibition is postulated to be an effective approach for treating metabolic syndrome.Using partially purified ACC from rat liver to screen herbs commonly used in Chinese folk medicine, we has found an ethanol extract of Polygonum hypoleucum Ohwi (EP) had potent ACC inhibitory activity.The metabolic benefits of EP were then tested on high glucose-stimulated HepG2 cells, high-fat (HF) diet-fed C57BL/6J mice and sucrose water-treated Wistar rats.Using this platform, some compounds isolated from EP were identified as ACC inhibitors.Our experiences show this screening platform, based on ACC inhibitory activity, can be used for developing Chinese nutraceuticals with potential in treating metabolic syndrome.
