  An alginate lyase gene,alga,was cloned from Abalone,encoding an endo-type polyM-specific alginate lyase AlgA.The molecule weight of the recombinant enzyme AlgA was about 30 KDa according to polyacrylamide gel electrophoresis (SDS-PAGE),after purified by Ni-NTA chro-matography,its specific activity was determinated to be 837.5 U/mg.The optimum temperature and pH of AlgA were 35 ℃ and 8 respectively,and it was a cold-adapted alginate lyase showing 50.6% and 80.5% of the maximal activity at 20 ℃ and 30 ℃ respectively.AlyA can only degrade poly mannuronate (polyM) but not poly guluronic acid (polyG),it can degrade polyM into unsaturated unsaturated dissaccharides,trisaccharides and tetrasaccharides.The AlgA was an endo-type polyM-specific alginate lyase.The Km and Vmax of AlgA toward polyM were 0.302 mg/ml and 1048.2 U/mg respectively.AlgA might be potential applications in alginate related biotechnology and industrial production.
