Purpose:Tovalidatethe quantificationofliversteatosisinvivobymDixonongeesewithwiderangeofliverfatcontent,using biochemicalextractiontechniqueasthereference. Methods:Twenty-twoLandesgeesewithoverfeedingfordifferentperiodwerecollected.MRquantificationofhepaticfat contentwereperformedonaclinical3Teslawhole-bodyMRIsystemforallgeesesamplesusingasix-echomDixonQuant gradient-echosequence.mDixonhepaticfatfractionmeasurementswereobtainedontheleft,right-superiorandright-inferior lobeforeachgooseliver.Forassessmentofintra-observeragreementofmDixonmeasurements,twoROImeasurementswere performedforallsamplesbythesameradiologistwithanintervalofonemonth.Thewholeliverwasextractedandcutintoleft lobe,right-superiorlobeandright-inferiorlobeforallstudiedgeese,andthecenterpartofeachlobewhichapproximately correspondedtothemDixonsamplingregionwascutdownforbiochemicalmeasurement(Soxhlet).Theintra-observer agreementofmDixonmeasurementswascalculatedusingBland-Altman95%limitsofagreementanalysis.Theaccuracyof mDixonfordeterminationoffatpercentagewasassessedthroughBland-Altman,usingSoxhletchemicalextractionasthegold standard,andPassingandBablokregressionequationwasthendetermined.Spearmencorrelationcoefficient(r)with95% confidenceintervals(CI)wascomputedtoexpressthedegreeoflinearassociationbetweenmeasures. Results:TheaveragefatpercentagedeterminedbymDixon(Fat%mDixon)andSoxhletchemicalextraction(Fat%Soxhlet)was 33.81±27.53%(range,0.88–79.41%)and24.13±21.1%(range,0.04–58.7%),respectively.MeandifferenceofthetwomDixon ROImeasurementsbythesameobserverwas0.1%(p=0.9914)witha95%CIof-3.7%–3.8%.mDixonwasdetectedhighly correlatedwithSoxhletchemicalextractionfordeterminationofliverfatpercentage,withaSpearmencorrelationcoefficientof 0.9487(p<0.001).mDixonoverestimatedliverfatpercentagecomparedtoSoxhletchemicalextractionwithameandifference(95%CI)betweenFat%mDixonandFat%Soxhletof9.68%(-5.28%–24.63%,p<0.001),andthedifferenceincreasedalongwiththe increaseofthevalueofliverfatpercentage(r=0.830,p<0.001).AccordingtoPassingandBablokregressionanalysis,Fat% SoxhletcouldbepredictedbyFat%mDixonwiththeequationas:Fat%Soxhlet=-1.8289+0.7728Fat%mDixon(ResidualStandard Deviation(RSD)±1.96SD:2.8819±5.6485%). Conclusion:Comparedtothebiochemicalextraction,mDixoncanbeusedforpredictingliverfatcontentinvivo.
