Successful incorporation of the paternal genome into the zygote represents one of the most fundamental requirements for maintaining genome integrity.During sperm maturation in animals, chromatin undergoes massive remodeling that replaces the bulk of chromosomal proteins, including histones, with protamines and similar proteins.This remodeling is believed to be essential for chromatin compaction in sperm nuclei.However, the molecular mechanisms governing this highly conserved process remain poorly understood.Upon fertilization, sperm chromatin undergoes de-condensation, in which paternal chromatin again undergoes extensive remodeling with the incorporation of maternally supplied factors.Little is known about this process as well.We have been studying two mutants in the fruit fly Drosophila that have defective incorporation of the paternal genome during fertilization, which inevitably results in haploid embryos maintaining only the maternal genome.Drosophila, with its amenability to genetic manipulations and its development program among the best characterized, would be one of the best system for the study of these two highly conserved processes of paternal genome remodeling.
