The genus Rhodococcus is a gram-positive high-G-C-content bacterium and exhibit organic solvent resistance as well as catalytic activities to degrade a large number of organic compounds.In particular,R.erythropolis can grow across a wide range of temperatures between 4+C and 35℃.We have selected R.erythropolis as a host cell and been aiming to create a multifunctional host that will be applicable to protein production along with a wide range of bioprocesses.For this purpose,we have isolated cryptic plasmids and insertion sequence from Rhodococcus and developed various expression vectors.Several self-replicable expression vectors and transposon-based integration vectors,which allows maintenance of expression cassettes without the use of antibiotics,were applicable to heterologous proteins expression under different environmental conditions.We also prepared a lysozyme-sensitive mutant cell that makes it easy to destroy its rigid cell wall and to recover the intracellular proteins.
