Rosiglitazone,a PPAR hiazolidinediones(TZDs) agonist,has been proposed to have neuroprotective effects in the central nervous system (CNS).However,the mechanisms underlying the beneficial effect of rosiglitazone are unclear.Live-cell imaging technology was used to investigate the effect of rosiglitazone on the dendritic filopodia and dendritic tree development and to find out the potential mechanism for the rosiglitazones effect.Results showed that rosiglitazone increased the dendritic filopodia density in a dose-dependent manner,but had no effect on the length and motility of dendritic filopodia.Neither the length nor the number of dendritic branches was altered by rosiglitazone.GW9662,an antagonist of PPARγ,completely blocked the increase of filopodia density mediated by rosiglitazone.These outcomes suggested that rosiglitazone may affect the initial stage of hippocampal neuron development through the PPAR pathway.This may be the underlying potential mechanism of the neuroprotective effect ofrosiglitazone.
