Taxanes (paclitaxel and docetaxel) are mitotic inhibitors and antimicrotubule agents that are highly active in the treatment of breast cancer.Both taxanes require solvents, Cremphor EL and polysorbate 80, respectively, to enter into solution.These solvents contribute to the main toxicities seen with taxanes, including severe hypersensitivity reactions.To avoid solvent-related adverse events, nab-paclitaxel, a solvent-free albumin-bound nanoparticle formulation of paclitaxel was developed and approved for treatment for metastatic breast cancer (MBC).nab-paclitaxel is not only safe, but also takes advantage of the increased delivery of albumin to tumors, and, thus, could deliver more paclitaxel to tumors than the solvent-based formulation.The absence of solvents and the albumin receptor-mediated delivery result in decreased toxicity and increased efficacy of nab-paclitaxel compared to solvent-based paclitaxel.
