Aim:α-taxilin,also known as IL14α,is a cytokine that identified and cloned from a Burkitt lymphoma cell line.The IL14 gene located in chromosome 4 in mouse and 1p34-6in human,where near the 1 p36 a lupus susceptibility locus.Recently,α-taxilin was identified as a novel binding partner of syntaxins.And it was found associated involved in intracellular vesicle traffic and in RCC metastatic,HCC proliferation.But its actual function in vivo is not yet known.In this moment,we aim to establish an animal model to investigate the actual function of α-taxilin,in vivo.Methods and materials:α-taxilin gene cloned from zebrafish was constructed in pTol2 vector.The reporter gene GFP and our object were promoted by CMV and EF1 α.Embryos were collected after fertilization,and then were injected with the p-Tol2-IL14-EGFP in one cell stage.Animals displaying EGFP expression at 48 hpf were selected and reared to adulthood.Founders with EGFP-positive were then outcrossed to AB fish.Heterozygotes were crossed to make homozygote,which were maintained as a stable line.Three lines were choose for the next analysis.The offspring from each line was collected to extract the genome DNA and whole protein.qPCR and western blotting were used to identified the RNA and protein expression profile in transgenic line.And histological chemical technologies were used to confirm its location in the whole mount and to observe the pathological chang in liver and kidney.Results:1.All the stable lines were confirmed that the α-taxilin were over-expressed in RNA and protein expression;2.IgM,IgD,IgT,Rag-1,TCRAC,IgLC-1,IgLC-2,IgLC-3 and Ikaros gene were examined using qPCR,the IgM was found significantly increased in transcription;3.The IgM was puried and seperate using SDS-PAGE followed by sliver staining,IgM was verified have a high expression in protein;4.And in the pathological part,a fibrosis in liver and decreased tubules were found.Conclusion:We constructed transgenic lines using zebrafish to analyse the role of α-taxilin in vivo,that have been verified is an available tool to study the function of α-taxilin.And we attempt to describe the physiological and pathological changes in it.From those,we know α-taxilin can disturb the immune system equilibrium,which may cause diseases.
