蛋白样品的荷电萃取电离质谱分析

近年来快速发展的常压电离质谱技术为复杂样品快速分析提供了一种新的高效策略.如何在无需复杂样品预处理的前提下,直接制备复杂基体样品中目标分子的离子是实际样品直接质谱分析的前提条件,而如何提高目标分子离子化效率和选择性则是该领域的关键科学问题.受液-液、液-固、液-气萃取分离技术的启发,课题组先后开发出多种独具特色用于分析复杂实际样品的常压离子化技术(电喷雾萃取电离(EESI)、表面解吸常压化学电离(DAPCI)、内部萃取电喷雾电离(iEESI)、气泡荷电萃取电离).微米级小气泡可选择性地携带带电溶液中的蛋白分子,通过气液萃取而与带电母体溶液分离,继而经去溶剂过程形成气相离子。将气体(如CO2、N2)导入充电蛋白溶液中,在微孔电极板上产生大量微米级气泡,蛋白分子会以气泡为载体,快速地从复杂基体溶液中分离,并带上一定量电荷,随后形成离子供质谱检测。实验研究了有机溶质的响应规律,并可以实现高盐溶液中生物蛋白的直接质谱分析。结果表明,气液荷电萃取电离技术对复杂样品溶液中有机溶质的富集和脱盐具有较好效果,能用于复杂样品的质谱分析领域。
