目的比较分析不同生长年限美花石斛茎组织构造及多糖含量。方法用植物组织切片技术和方法观察比较美花石斛组织构造差异和变化;用硫酸-苯酚法测定多糖含量。结果不同生长年限美花石斛茎的表皮、基本组织以及维管束差异比较大;1年生、2年生和3年生美花石斛茎多糖含量分别为18.36%、23.60%、15.42%。结论不同生长年限的美花石斛组织构造特征和差异能够反应“表面金黄色、质地柔软和嚼之黏性”等传统品质评价指标;采收期安排在第2年的生长期结束时比较合适,至于具体采收时间,还需要进一步动态分析和研究才能确定。
Objective To compare and analyze the stem tissue structure and polysaccharide content of Dendrobium nobile in different growth years. Methods Plant tissue sectioning techniques and methods were used to observe and compare the structural differences and changes of Dendrobium nobile. The content of polysaccharides was determined by sulfuric acid-phenol method. Results The stems, basic tissues and vascular bundles of Dendrobium candidum were different in different growth years. The contents of polysaccharides in stems of 1st, 2nd and 3rd years were 18.36%, 23.60% and 15.42%, respectively. Conclusion The structural characteristics and differences of different flowering ages of Dendrobium can reflect the traditional quality evaluation indexes such as “golden surface, soft texture and stickiness”, and the harvesting period is suitable for the end of the growth period of the second year. As for the specific harvest time, it needs further dynamic analysis and research to determine.
