The prefrontal cortex (PFC) plays a critical role in cognitive functions such as working memory,attention regulation and response inhibition.Extensive behavioral and pharmacological studies in rodents, monkeys and humans have shown that stimulation of prefrontal cortical α2A-adrenoceptors (α2A-ARs) improves working memory.However, expression of PFC α2A-ARs, especially in rodent, is largely unknown.Here, we investigated the expression of t2A-ARs in pyramidal neurons, interneurons (PV-, CR-and CB-immunoreactive interneurons)as well as presynaptic terminals of rat PFC using double-labeling immunofluorencence technique.Our results showed that α2A-ARs expressed in the majority of pyramidal neurons and extensively expressed in interneurons.Furthermore, α2A-ARs-expressing pyramidal neurons uniformly distributed throughout different layers of the mPFC, while α2A-ARs-expressing interneurons were differentially distributed in layer 5-6 than layer 2-3.α2AARs are co-expressed with vesicular glutamate transporterl (vGluT1) and glutamic acid decarboxylase (GAD65),suggesting that α2A-ARs are present on the glutamatergic terminals and GABAergic terminals of mPFC.
