目的体外观察红花多糖(SPS)对人胃癌SGC-7901细胞线粒体膜电位(MMP)及细胞增殖的影响,初步探讨SPS对SGC-7901细胞的抑制机制。方法不同浓度的SPS处理SGC-7901细胞,倒置显微镜观察SGC-7901细胞的形态学变化,透射电镜观察SGC-7901细胞超微形态结构变化,罗丹明123染色观察SGC-7901细胞MMP变化,MTT比色法检测SGC-7901细胞的增殖。结果倒置显微镜观察到经SPS处理的贴壁细胞体积变小,部分细胞崩解,有凋亡小体形成,随剂量增加愈加明显;透射电镜观察到经SPS处理的细胞线粒体肿胀、胞核浓缩偏移,呈凋亡形态改变;罗丹明123染色可见经SPS处理的细胞内荧光强度显著下降,该作用呈明显的时间依赖性;MTT检测显示经SPS处理的细胞体外增殖受抑制,该作用呈明显的剂量依赖性。结论 SPS可以降低人胃癌SGC-7901细胞MMP,同时能明显抑制SGC-7901细胞体外增殖并可诱导细胞凋亡,该作用呈一定的时间与剂量依赖。
Objective To observe the effects of safflower polysaccharide (SPS) on mitochondrial membrane potential (MMP) and cell proliferation of human gastric cancer cell line SGC-7901 in vitro and to investigate the inhibitory mechanism of SPS on SGC-7901 cells. Methods SGC-7901 cells were treated with different concentrations of SPS. The morphological changes of SGC-7901 cells were observed under inverted microscope. The ultrastructure of SGC-7901 cells was observed by transmission electron microscope. The changes of MMPs and SGT were observed by rhodamine 123 staining Colorimetric assay of proliferation of SGC-7901 cells. Results The inverted microscope showed that the size of the adherent cells decreased, some cells disintegrated and apoptotic bodies formed, which became more obvious with the increase of dose. The mitochondria swelling and the concentration of nuclei in the SPS treated cells were observed by transmission electron microscope The apoptotic morphological changes were observed. Rhodamine123 staining showed that the intracellular fluorescence intensity decreased markedly by SPS, which showed a significant time-dependent effect. MTT assay showed that the proliferation of SPS-treated cells was inhibited in vitro Dose-dependent. Conclusion SPS can reduce the MMP of human gastric cancer cell line SGC-7901, at the same time it can significantly inhibit SGC-7901 cell proliferation in vitro and induce apoptosis, which is time-dependent and dose-dependent.
