目的对PfCP-2.9/PfCSP-2疟疾联合疫苗所诱发的特异性IgG抗体进行分析评价。方法将5只新西兰白兔分为两组,其中免疫组3只,非免疫组2只。免疫组接种PfCP-2.9/PfCSP-2疟疾联合疫苗,非免疫组注射生理盐水。连续3次免疫后,采集血液样本并经间接ELISA检测其IgG抗体水平。根据非免疫组兔的吸光度数值(A值)计算临界值。临界值=2.1×非疫苗免疫组兔A值均值。若免疫组兔吸光度值高于临界值,则被判定为疫苗可诱发特异性IgG抗体。结果疫苗可诱发针对PfCP-2.9和PfCSP-2重组蛋白IgG抗体,经4个稀释浓度(1:100、1:200、1:400、1:800)检测获得2只免疫组兔A值分别为0.629/0.544、0.548/0.443、0.479/0.378、0.410/0.281,均高于相应稀释浓度的临界值(0.019、0.017、0.016、0.020);疫苗可诱发针对PfCSP的特异性IgG抗体(每孔包被4μg/ml多肽),经4个稀释浓度(1:100、1:200、1:400、1:800)检测2只免疫组家兔A值分别为0.308/0.152、0.194/0.080、0.110/0.040、0.060/0.025,均高于相应稀释浓度的临界值(0,027、0.019、0,018、0.018);疫苗可诱发针对PfCSP的特异性IgG抗体(每孔包被10μg/ml多肽),经4个稀释浓度(1:100、1:200、1:400、1:800)检测获得2只免疫组家兔A值分别为0.333/0.129、0.22I/0.086、0.133/0.048、0.075/0.029,均高于相应稀释浓度的临界值(0.048、0.038、0.036、0.036);结论PfCP-2.9/PfCSP-2疟疾联合疫苗可诱发高水平IgG抗体,该疫苗是安全、有效且耐受性良好。
Objective To analyze and evaluate the specific IgG antibodies induced by the PfCP-2.9 / PfCSP-2 malaria vaccine. Methods Five New Zealand white rabbits were divided into two groups, of which 3 were immunized group and 2 were non-immunized group. The vaccine induced specific IgG antibody against PfCSP (10μg / ml polypeptide per well) (1: 100, 1: 200, 1: 400 and 1: 800), the A values of the two immunized rabbits were 0.333 / 0.129,0.22I / 0.086,0.133 / 0.048,0.075 / 0.029 respectively, (0.048,0.038,0.036,0.036). Conclusion The combination of PfCP-2.9 / PfCSP-2 malaria vaccine can induce high level of IgG antibody. The vaccine is safe, effective and well tolerated.
