目的:采用反相HPLC建立何首乌提取液益生菌发酵前后指纹图谱,观察何首乌提取液发酵前后化学成分变化与活性的相关性。方法:以Agilent ZORBAX SB-AQ(4.6 mm×150 mm,5.0μm)为色谱柱,以A(乙腈)和B(0.1%磷酸)为流动相梯度洗脱,流速0.8 mL·min-1,柱温20℃,检测波长254 nm。结果:建立了有13个共有峰的HPLC色谱图,并比较了色谱峰相对峰面积的变化。结论:何首乌提取液经益生菌发酵后化学成分的变化较大,中药经过发酵可通过多途径影响化学成分,进而改变药物的药理药效。
OBJECTIVE: To establish a fingerprint chromatogram of probiotics of Polygonum multiflorum Thunb before and after fermentation by reversed-phase HPLC, and to observe the correlation between chemical composition changes and activity before and after the fermentation of Polygonum multiflorum Thunb extract. Methods: The mobile phase consisted of Agilent ZORBAX SB-AQ column (4.6 mm × 150 mm, 5.0 μm), gradient elution with A (acetonitrile) and B (0.1% phosphoric acid) at a flow rate of 0.8 mL · min -1 Temperature 20 ℃, detection wavelength 254 nm. Results: HPLC chromatograms of 13 common peaks were established and the relative peak areas of the peaks were compared. CONCLUSION: The chemical composition of Polygonum multiflorum Thunb extract after fermentation by probiotics is quite different. After fermentation, traditional Chinese medicine can affect the chemical composition through many channels and thus change the pharmacological effects of the drug.
