目的考察乳腺癌组织(Ca)及癌旁正常乳腺组织(Nor)中胸腺嘧啶磷酸化酶(TP酶)及二氢嘧啶脱氢酶(DPD酶)与化疗药物卡培他滨(CAP)及其活性代谢产物氟尿嘧啶(5-Fu)浓度的相关性。方法应用ELISA方法定量测定26例经病理证实为乳腺癌患者的癌组织及正常组织中TP酶及DPD酶的表达水平;应用高效液相色谱法(HPLC)定量测定癌组织及正常组织中的CAP及5-Fu浓度。结果TP酶在癌组织中平均表达水平是386.1163ng/mg蛋白,在正常组织中为42.5682ng/mg蛋白。DPD酶在癌组织中平均表达水平是209.528ng/mg蛋白,在正常组织中为339.368ng/mg蛋白。CAP/5-Fu比值范围是0.0804~5.0000。结论在P=0.05水平上,癌组织中TP酶表达水平远远高于其在正常组织中的表达水平,DPD酶在癌组织及正常组织中的表达水平没有差异,但具有很强的相关性(Spearman相关系数=0.43>rs0.05/2(26)=0.390,P<0.05)。TP/DPD预测CAP/5-Fu的效能高于TP酶及DPD酶。
Objective To investigate the relationship between thymidine phosphorylase (TPase), dihydropyrimidine dehydrogenase (DPDase) and chemotherapeutic drug capecitabine (CAP) in breast cancer tissue (Ca) and adjacent normal breast tissue Correlation of active metabolite 5-Fu concentrations.
