目的探讨督脉电针与神经干细胞移植联合应用对脊髓全横断大鼠受损伤的神经元存活及其轴突再生的影响。方法将对照组、神经干细胞移植组、督脉电针组和督脉电针+神经干细胞移植组的成年大鼠胸10脊髓段做全横断损伤;其中神经干细胞移植组和电针神经干细胞组在损伤处移植神经干细胞。电针组和电针神经干细胞移植组在术后开始接受督脉电针治疗。所有动物存活67d。结果1.电针神经干细胞移植组脊髓损伤处的去甲肾上腺素能5、_羟色胺受体、降钙素基因相关肽能和生长相关蛋白-43阳性染色的4种神经纤维均明显多于其他几组。2.电镜下可观察到脊髓横断处有再生的神经纤维穿越,在电针神经干细胞移植组尤为明显。3.大脑体感运动区皮质和中脑红核受损伤的神经元存活数量也多于其他几组,一些神经元的再生神经纤维可能穿越横断处,进入尾端脊髓组织。结论督脉电针与神经干细胞移植联合应用能促进脊髓全横断大鼠受损伤的神经元存活及其轴突再生。
Objective To investigate the effects of combination of Du meridian electroacupuncture and neural stem cell transplantation on neuronal survival and axon regeneration in injured spinal cord transected rats. Methods Thoracic 10 spinal cord segments of adult rats in the control group, neural stem cell transplantation group, Du Mai electroacupuncture group and Du Fu electroacupuncture plus neural stem cell transplantation group were transected completely. Among them, neural stem cell transplantation group and EA group Injured at the transplantation of neural stem cells. Electroacupuncture and electroacupuncture of neural stem cells transplantation group began to receive Du meridian acupuncture after operation. All animals survived 67d. Results1.Nernergic fibers of norepinephrine, serotonin receptor, calcitonin gene-related peptide and growth-associated protein-43 positive staining in spinal cord injury of EA group were significantly more than those in other groups Several groups. Electron microscopy can be found in the transected spinal cord regenerated nerve fiber traversing, especially in EA group. Neurons in the somatosensory motor cortex and midbrain red nucleus survived more than those in other groups. The regenerated nerve fibers of some neurons may cross the transection and enter the caudal spinal cord tissue. Conclusion The combination of Du meridian electroacupuncture and neural stem cell transplantation can promote the survival of injured neurons and their axon regeneration in rats with spinal cord transection.
