目的:从炎性反应和氧化应激两方面探讨复方苦参汤对溃疡性结肠炎(UC)的治疗作用。方法:雄性SD大鼠48只,随机分为正常组,模型组,美沙拉秦组、复方苦参汤低、中、高剂量组,除正常组外,各组均以2,4,6-三硝基苯磺酸灌肠法建立UC模型。相应各组分别予以美沙拉秦混悬液,低、中、高剂量复方苦参汤灌胃,每天1次,第8天取结肠组织检测IL-1β、TNF-α、IL-10、MDA、SOD的表达。结果:与模型组比较,美沙拉秦组及复方苦参汤高剂量组大鼠疾病活动指数,结肠组织宏观损伤评分和组织病理学均得到明显改善,并且结肠IL-1β、TNF-α和MDA表达明显下调(P<0.05),IL-10和SOD含量明显升高,且呈剂量依赖性。结论:复方苦参汤对UC的治疗作用与降低促炎因子和氧化介质的表达,升高抑炎因子和抗氧化介质的表达有关。
Objective: To investigate the therapeutic effect of compound Kushen decoction on ulcerative colitis (UC) from the aspects of inflammatory reaction and oxidative stress. Methods: Forty-eight male Sprague-Dawley rats were randomly divided into normal group, model group, mesalamine group, compound Kushen Tang low, medium and high dose group, except the normal group, Trinitrobenzene sulfonic acid enema to establish UC model. The corresponding groups were given mesalazine suspension, low, medium and high doses of Compound Kushen Tang gavage once a day, on the 8th day colonic tissue was taken to detect IL-1β, TNF-α, IL-10, MDA, SOD expression. Results: Compared with the model group, the disease activity index, macroscopic damage score and histopathology of the rats in the mesalazine group and the compound Kushen Tang high-dose group were significantly improved, and the levels of IL-1β, TNF-α and MDA (P <0.05). The levels of IL-10 and SOD were significantly increased in a dose-dependent manner. Conclusion: The therapeutic effect of Fufang Kushen decoction on UC is related to decreasing the expression of proinflammatory cytokines and oxidative mediators, increasing the expression of anti-inflammatory cytokines and antioxidants.
