以正丁酸和巴豆油诱导的Raji细胞为免疫原,应用杂交瘤技术制备一种抗EB病毒(EBV)早期R型抗原(EA-R)的鼠IgM单克隆抗体,命名为BAE-5。以BAE-5为一抗应用APAAP免疫组织化学技术检测34例鼻咽癌和29例非鼻咽癌,结果表明,所有的鼻咽癌(34例)和29例非鼻咽癌肿瘤中的9例能与BAE-5显阳性反应。与已进行EB病毒DNA原位杂交的34例鼻咽癌结果对比认为:大多数鼻咽癌细胞不仅可检测出EBV-DNA,而且这些细胞中的一部分还可表达EA-R。说明这些细胞中的EBV-DNA处于激活状态。一些非鼻咽癌肿瘤能与BAE-5起阳性反应,可能是由于这些肿瘤细胞中也有EB病毒早期抗原存在或者可能存在与EA-R抗原决定簇相类似结构的多肽而引起交叉反应。作者认为这些结果对了解EB病毒有关的多肽,特别是EA复合物在鼻咽癌和一些含EBV-DNA的非鼻咽癌肿瘤的发生和发展中的作用具有理论意义。
Using Raji cells induced by n-butyric acid and croton oil as immunogens, a monoclonal antibody against murine IgM against Epstein-Barr virus (EBV) early type R antigen (EA-R) was prepared using the hybridoma technique, named BAE-5. Using BAE-5 as the primary antibody, APAAP immunohistochemistry was used to detect 34 cases of nasopharyngeal carcinoma and 29 cases of non-nasopharyngeal carcinoma. The results showed that all of the nasopharyngeal carcinoma (34 cases) and 29 cases of non-nasopharyngeal carcinoma Cases were positive with BAE-5. Compared with the results of 34 cases of nasopharyngeal carcinoma that have undergone in situ hybridization of Epstein-Barr virus DNA, most nasopharyngeal carcinoma cells can not only detect EBV-DNA, but also some of these cells can express EA-R. This shows that the EBV-DNA in these cells is active. Some non-nasopharyngeal cancers can react positively with BAE-5, possibly due to the presence of EBV early antigens in these tumor cells or the possible presence of a polypeptide with a structure similar to that of the EA-R epitope. The authors believe that these results are of theoretical significance for understanding the role of EB virus-related peptides, particularly EA complexes, in the development and progression of nasopharyngeal carcinoma and some non-nasopharyngeal cancers containing EBV-DNA.
