目的探讨研究不同种类的增菌培养基对于沙门菌的增菌效果,并进行菌株分离效果的比较与分析。方法选择目前市面上较为常用的四硫磺酸盐增菌液(Tetrathionate Broth Base,TTB)、亚硒酸盐胱氨酸增菌液(Selenite Cystine Borth,SC)、罗伯特增菌液(Rappaport-Vassiliadis Soya Broth,RVS)和亚硒酸盐煌绿增菌液(Selenite Brilliant Green Broth,SBG)4种沙门菌选择性增菌培养基作为研究对象,首先采用沙门菌和大肠埃希氏菌标准菌株进行增菌培养,观察增菌效果;然后采用上述4种增菌培养基对健康人群标本和食品标本进行检测,观察和比较增菌效果。结果对鼠伤寒沙门菌、肠炎沙门菌、猪霍乱沙门菌和伤寒沙门菌的24 h增菌动态观察效果表明,SBG增菌培养基能够在最短时间内使4种沙门菌达到对数生长期,且稳定期菌株的OD值均高于其它3类培养基,差异有统计学意义(χ2=8.57,P<0.05),并且能够有效抑制大肠埃希氏菌的生长;对健康人群的肛拭子沙门菌分离率是0.2%,对食品的分离率是13.5%,均高于其它增菌液效果。结论 SBG选择性增菌培养基是目前市售的对沙门菌增菌效果最佳的培养基之一,采用高效的增菌培养基和分离培养基联合应用能够大幅度提高沙门菌的检出率。
Objective To study the effects of different kinds of enrichment medium on the enrichment of Salmonella, and to compare and analyze the isolation effect of strains. Methods Tetrathionate Broth Base (TTB), Selenite Cystine Borth (SC), Rappaport-Vassiliadis Soya Broth, RVS) and selenite Brilliant Green Broth (SBG) were selected as the research object. Firstly, the standard strains of Salmonella and Escherichia coli Bacterial culture, to observe the effect of enrichment; and then use the above four kinds of enrichment medium samples and healthy food samples were tested to observe and compare the effect of enrichment. Results The dynamic observation results of 24 h enrichment of Salmonella typhimurium, Salmonella enteritidis, Salmonella choleraesuis and Salmonella typhi showed that SBG enrichment medium can reach the logarithmic growth phase of the four Salmonella species in the shortest time, And the OD values of the stable strains were higher than those of the other three kinds of media, the difference was statistically significant (χ2 = 8.57, P <0.05), and can effectively inhibit the growth of Escherichia coli; for the healthy population anal swab The isolation rate of Salmonella was 0.2% and the isolation rate of food was 13.5%, which were higher than those of other bacteriostasis. Conclusion SBG selective enrichment medium is one of the most commercially available commercially available Salmonella enrichment broths. The combination of efficient enrichment medium and isolation medium can greatly improve the detection rate of Salmonella .
