[目的]探讨肿瘤抑制基因对肺腺癌细胞生长的抑制作用。[方法]用FuGene转染方法将p16的表达质粒分别转入两个p16未表达的具高转移能力的肺腺癌细胞系A2和A549中 ,并对转染前后的肿瘤细胞进行了细胞生长曲线、凋亡检测、电镜和流式细胞仪分析。[结果]发现A2、A549细胞系转染后的细胞生长曲线的斜率要比转染前低得多 ,电镜分析也可看出转染前后的细胞形态发生了改变 ,但均未检测到凋亡信号。经流式细胞仪分析发现 :转染后的细胞系均在G0~G1 期发生阻滞。[结论]p16基因在A2、A549细胞中表达可以抑制其生长。对于发生了p16基因突变或缺失而导致p16蛋白在数量或功能上发生改变的恶性肿瘤 ,外源性p16基因的导入不失为一种治疗恶性肿瘤的有效途径
[Objective] To investigate the inhibitory effect of tumor suppressor gene on the growth of lung adenocarcinoma cells. [Method] The expression plasmids of p16 were transfected into two non-expressing high-metastatic lung adenocarcinoma cell lines A2 and A549 respectively by FuGene transfection method. The cell growth curves of the tumor cells before and after transfection , Apoptosis detection, electron microscopy and flow cytometry analysis. [Result] It was found that the slope of cell growth curve after transfection of A2 and A549 cell lines was much lower than that before transfection. The morphological changes of cells before and after transfection were also observed by electron microscopy, but no apoptosis was detected signal. Flow cytometry analysis found: transfected cell lines in the G0 ~ G1 block. [Conclusion] The expression of p16 gene in A2 and A549 cells can inhibit its growth. Introduction of exogenous p16 gene is an effective approach to the treatment of malignant tumors in malignant tumors in which mutations or deletions of p16 gene result in the change of quantity or function of p16 protein
