目的:探讨甘麦大枣汤加味方对抑郁症模型大鼠海马信号转导cAMP-蛋白激酶A(PKA)途径的影响。方法:以孤养加慢性不可预见性中等刺激所致抑郁症大鼠为模型,灌服甘麦大枣汤加味方,通过旷场行为测定、糖水消耗试验观察行为变化,放免法测定海马cAMP含量以及原位杂交法测定海马5-HT1A受体、PKAmRNA表达。结果:模型组大鼠垂直活动、水平运动及糖水消耗显著下降,海马cAMP、PKAmRNA表达显著上升(P<0.01)。用药组与模型组比较,水平运动及垂直活动上升,糖水消耗增加,cAMP含量显著下降,PKAmRNA表达显著下降(P<0.01)且呈量效关系。结论:甘麦大枣汤加味方下调抑郁症大鼠海马信号转导cAMP-PKA途径可能是该方纠正抑郁症模型大鼠行为学变化的环节之一。
Objective: To investigate the effect of Ganmai Dazao Decoction on the signal transduction of cAMP-protein kinase A (PKA) pathway in hippocampus of depressive model rats. METHODS: Rats with depressive disorder caused by isolation and chronic unpredictable moderate stimulation were fed with Ganmai Dazao Decoction. The behavioral changes were observed by behavioral measurements of the open field, sugar consumption test, and hippocampal cAMP content was determined by radioimmunoassay. The expression of 5-HT1A receptor and PKA mRNA in hippocampus was determined by in situ hybridization. RESULTS: Vertical activity, horizontal exercise, and sugar consumption were significantly decreased in the model group, and cAMP and PKA mRNA expression in the hippocampus significantly increased (P<0.01). Compared with the model group, the medication group increased horizontal exercise and vertical activity, increased the consumption of sugar and water, markedly decreased cAMP content, and significantly decreased the expression of PKA mRNA (P<0.01). Conclusion: Ganmai Dazao Decoction can down-regulate the cAMP-PKA pathway in the hippocampus of depressive rats, which may be one of the links in correcting the behavioral changes of depressive model rats.
