目的建立用于测定肌萎缩侧索硬化(ALS)患者脑脊液中氨基酸类神经递质的方法,观察患者用肌萎灵注射液治疗前后脑脊液中氨基酸类神经递质的变化。方法采集42例患者的脑脊液,用邻苯二甲醛(OPA)柱前衍生变增益HPLC法测定脑脊液中天冬氨酸、谷氨酸、甘氨酸、γ-氨基丁酸的含量。结果4种氨基酸在20min内分离度、线性关系均良好;42例ALS患者治疗后与治疗前相比,脑脊液中兴奋性的天门冬氨酸和谷氨酸水平明显降低,而抑制性的甘氨酸和γ-氨基丁酸水平变化不明显。结论本方法快速、准确、灵敏度高,适用于脑脊液中神经递质类氨基酸的含量测定。
Objective To establish a method for determination of amino acid neurotransmitters in cerebrospinal fluid of patients with amyotrophic lateral sclerosis (ALS) and to observe the changes of amino acid neurotransmitters in cerebrospinal fluid in patients before and after treatment with muscle wilting injection. Methods Cerebrospinal fluid was collected from 42 patients. The content of aspartic acid, glutamic acid, glycine, and γ-aminobutyric acid in cerebrospinal fluid was determined by pre-column derivatization with modified phthalazine (OPA). Results The four amino acids showed good linearity within 20min. The levels of excitatory aspartate and glutamate in cerebrospinal fluid in 42 patients with ALS were significantly lower than those before treatment, while the inhibitory glycine and γ-aminobutyric acid levels did not change significantly. Conclusion The method is rapid, accurate and sensitive. It is suitable for the determination of neurotransmitter amino acids in cerebrospinal fluid.
