A study of genetic polymorphism of Toll-like receptor 4 at promoter region in Zhuang and Han population in Guangxi

【Abstract】 Objective To investigate the distribution of polymorphisms of rs10116253C/T and rs10983755G/A loci of Toll-like receptor 4(TLR4) at promoter region in Zhuang and Han population in Guangxi,and to explore their differences among various regions and groups.
Methods Snapshot SNPs genotyping assays and DNA sequencing methods were used to detect genotype and allele of rs10116253C/T and rs10983755G/A loci in TLR4 gene promoter region among 123 healthy cases in Zhuang and Han population in Guangxi,and the distribution difference of genotype and allele frequency between Hapmap-HCB,Hapmap-JPT,Hapmap-CEU and Hapmap-YRI was compared and analyzed.
Results Three genotypes(CC,CT and TT) were found in rs10116253C/T of TLR4 gene at promoter region in Zhuang and Han population in Guangxi with the frequency of 13.82%,44.72% and 41.46% respectively,difference of the three genotypes was statistically significant between genders in Guangxi population(P<0.05).However,there was no statistically significant difference in each allele between different genders in Guangxi population(P>0.05).Difference of genotype and allele frequencies of rs10116253C/T between Guangxi population and Hapmap-CEU was statistically significant(P<0.05).Three genotypes(AA,AG and GG) were found in rs10983755G/A with the frequency of 4.07%,32.52% and 63.41%,respectively.There was no statistically significant difference in each genotype and allele between different genders in Guangxi population(P>0.05),but difference of genotype and allele distribution frequencies rs10983755G/A between Guangxi population and Hapmap-CEU and Hapmap-YRI was statistically significant(P<0.001).
Conclusion The polymorphisms of TLR4 gene rs10116253C/T and rs10983755G/A at promoter region are different in various regions and ethnic groups.

Toll样受体(Toll-like receptors,TLRs)蛋白被确认为基因家族中相对保守的跨膜受体之一,属于Ⅰ型跨膜糖蛋白受体,是人类天然性免疫家族中的基本成員之一,并在免疫调节及炎性反应过程中发挥关键作用[1~2]。随着分子生物学水平的进步和基因组学技术的研究,作为TLRs家族中重要的成员之一的Toll样受体4(Toll-like receptor 4,TLR4),其多态性对疾病的影响也逐渐受到国内外学者们的关注,多项研究证实TLR4基因多个位点(如rs11536889、rs4986790、rs4986791、rs1927911、rs10759932等)具有多态性,与感染[3]、肿瘤[4]、自身免疫性疾病[5]、冠心病[6]等多种疾病的易感性及预后密切相关。但基因多态性受地区、人种及环境等方面因素的影响,故本研究拟探讨广西壮族和汉族健康人群TLR4基因启动子区rs10116253C/T、rs10983755G/A位点单核苷酸多态性(Single nucleotide polymorphism,SNP)的分布特点,同时与不同地区、不同种族人群比较,旨在分析基因多态性在不同地区、不同种族中的分布差异,为与TLR4基因有关疾病的遗传学规律及治疗等方面的研究提供参考依据。
