目的:研究姜黄素在小鼠内毒素血症发病中起的作用及意义。方法:选用50只清洁级ICR小鼠随机均分为5组,每组10只。5组分别为:预防剂量组、治疗剂量组、药物对照组、空白对照组。于攻毒前和攻毒后6、12、18和24 h检测各组小鼠的直肠温度,给药5天后分别观察小鼠的死亡率、病理解剖观察心、肝、肾和肠病变,眼球取血进行白细胞、红细胞、血小板、血红蛋白测定,分离血清,用固相夹心法酶联免疫吸附法测定血清TNF-α、IL-1、IL-6水平。结果:预防剂量组、治疗剂量组小鼠较LPS对照组小鼠死亡率下降,在6 h和12 h时,治疗剂量组和预防剂量组均显著或极显著高于LPS对照组(P<0.05或P<0.01),治疗组和预防组的白细胞、血小板数明显升高,差异显著(P<0.05)。治疗组、预防组与LPS组相比,差异显著(P<0.01)。治疗组、预防组的TNF-α、IL-1、IL-6水平较LPS组低,前者与正常组有显著性差异(P<0.05),后者与正常组没有显著性差异(P>0.05)。结论:姜黄素对内毒素血症小鼠有保护作用,预防、治疗均有效果,预防效应强于治疗。
Objective: To study the role and significance of curcumin in the pathogenesis of mouse endotoxemia. Methods: 50 clean ICR mice were randomly divided into 5 groups with 10 mice in each group. Five groups were: preventive dose group, therapeutic dose group, drug control group, blank control group. The rectal temperature of mice in each group was detected before challenge and at 6, 12, 18 and 24 h after challenge. The mortality of mice was observed 5 days after the challenge. Pathological anatomy was used to observe the changes of heart, liver, kidney and intestine, Blood was collected for determination of leucocyte, erythrocyte, platelet and hemoglobin, serum was separated and serum TNF-α, IL-1 and IL-6 levels were measured by solid-phase sandwich enzyme-linked immunosorbent assay. Results: Compared with the LPS control group, the mortality of mice in the preventive dose group and the therapeutic dose group was significantly lower than that of the LPS control group (P <0.05) at 6 h and 12 h Or P <0.01). The number of white blood cells and platelets in treatment group and prevention group were significantly increased (P <0.05). Compared with LPS group, the difference between the treatment group and the prevention group was significant (P <0.01). The levels of TNF-α, IL-1 and IL-6 in the treatment group and the prevention group were lower than those in the LPS group (P <0.05), while there was no significant difference between the latter group and the normal group (P> 0.05 ). Conclusion: Curcumin has a protective effect on endotoxemia mice, prevention and treatment are effective, prevention is stronger than treatment.
