目的:探讨清热化瘀方联合脑缺血预处理对大鼠脑缺血再灌注损伤的神经保护作用。方法:健康雄性SD大鼠160只,随机分为4组:假手术组、大脑中动脉缺血组(middle cerebral artery occlusion,MCAO)、脑缺血预处理组(brain ischemic preconditioning,BIP)、清热化瘀方组(Qingre Huayu prescription,QRHY),每组按照再灌注后12 h,1,2,3 d 4个时间点平均分为4个亚组,制备缺血预处理模型,评价脑缺血预处理对缺血再灌注大鼠脑组织病理形态学、脑梗死体积和神经细胞凋亡的影响及清热化瘀方的干预作用。结果:①光镜和电镜下观察到BIP组神经细胞的病理形态学改变较MCAO组明显减轻,且BIP组梗死面积明显小于MCAO组(P<0.05),QRHY组较BIP组上述损害进一步减轻(P<0.05)。②大鼠脑缺血再灌注后12 h,MCAO组细胞凋亡发生率较假手术组显著增加(P<0.01),1 d时达到高峰,以后时间点逐渐下降,但仍高于假手术组(P<0.01);BIP组各个时间点神经元凋亡发生率较MCAO组显著降低(P<0.05或P<0.01);QRHY组较BIP组神经细胞凋亡率进一步降低(P<0.05或P<0.01)。结论:清热化瘀方联合缺血预处理对大鼠脑缺血再灌注损伤有协同保护作用。
Objective: To explore the neuroprotective effects of Qingre Huayu Recipe combined with cerebral ischemic preconditioning on cerebral ischemia-reperfusion injury in rats. Methods: 160 healthy male SD rats were randomly divided into 4 groups: sham operation group, middle cerebral artery occlusion (MCAO) group, brain ischemic preconditioning group (BIP) The Qingre Huayu prescription (QRHY) group was divided equally into 4 subgroups at 12 h, 1, 2 and 3 d after reperfusion, and the ischemic preconditioning model was established to evaluate the effect of cerebral ischemia Effects of Preconditioning on Pathomorphology, Cerebral Infarction Volume and Neuronal Apoptosis in Rats with Ischemia / Reperfusion and the Intervening Effect of “Qingre Huayu Decoction”. Results: ①The pathological changes of BIP neurons in the BIP group were significantly lower than those in the MCAO group (P <0.05), and the infarct size in the BIP group was significantly lower than that in the MCAO group (P <0.05) P <0.05). ②The apoptosis rate of MCAO group was significantly higher than that of sham-operation group (P <0.01) at 12 h after cerebral ischemia-reperfusion, and peaked at 1 d, then gradually decreased but remained higher than that of sham operation group (P <0.01). The apoptosis rate of neurons in BIP group was significantly lower than that in MCAO group at each time point (P <0.05 or P <0.01), and apoptosis rate of neurons in QRIP group was lower than that of BIP group (P <0.05 or P <0.01). Conclusion: Qingre Huayu Recipe combined with ischemic preconditioning has a synergistic protective effect on cerebral ischemia-reperfusion injury in rats.
