目的:探讨白细胞介素-lβ(IL-lβ)和肿瘤坏死因子-α(TNF-α)对牙周膜细胞内白血病抑制因子(LIF)表达的影响。方法:体外分离培养鉴定人牙周韧带细胞(HPDLC),取第3代细胞用于实验,利用牙周改建中高表达因子TNF-α和IL-1β刺激HPDLC后,实时定量反转录-聚合酶链反应检测LIF mRNA的表达。结果:IL-1β在浓度为0.1ng/mL和5 ng/mL时,均显著促进LIF的分泌(P<0.01)。TNF-α在浓度为10 ng/mL时,明显促进LIF的分泌(P<0.05)。结论:牙周改建中高表达细胞因子IL-1β和TNF-α可促进牙周膜细胞中LIF的表达。
Objective: To investigate the effect of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) on the expression of leukemia inhibitory factor (LIF) in periodontal ligament cells. METHODS: Human periodontal ligament cells (HPDLC) were isolated and cultured in vitro. The third passage cells were used for the experiment. HPDLC was stimulated by high expression of TNF-α and IL-1β during periodontal reconstruction. Response to detect LIF mRNA expression. Results: IL-1β significantly promoted the secretion of LIF (P <0.01) at the concentrations of 0.1 ng / mL and 5 ng / mL. TNF-α at a concentration of 10 ng / mL, significantly promote the secretion of LIF (P <0.05). CONCLUSION: High expression of cytokines IL-1β and TNF-α during periodontal reconstruction can promote the expression of LIF in periodontal ligament cells.
