目的了解2010-2013年深圳市南山区环境中不同样品中分离的创伤弧菌质粒携带情况及基因的遗传多样性。方法采用聚合酶链反应技术检测深圳市南山区环境样品中创伤弧菌的6种质粒基因:seq25、seq51、Tra I、Tra I2、Tra D及Vep07。采用多位点序列分子分型方法进行分子分型及聚类分析。结果 50株创伤弧菌中,43株菌(43/50,86%)携带质粒,形成19种质粒图谱:20株(20/50,40%)同时存在毒力与结合质粒,13株(13/50,26%)仅存在毒力质粒,2株(2/50,4%)仅存在结合质粒。多位点序列分型方法将50株创伤弧菌,分为43个ST型。结论质粒广泛存在于深圳市南山区环境样品分离的创伤弧菌中,不同类型样本分离菌株携带质粒存在差异且具有遗传多样性。
Objective To understand the plasmid carrying status of Vibrio vulnificus isolated from different samples in Nanshan District of Shenzhen City in 2010-2013 and its genetic diversity. Methods Polymerase chain reaction (PCR) was used to detect six kinds of plasmids of Vibrio vulnificus in sera of Shenzhen Nanshan District: seq25, seq51, Tra I, Tra I2, Tra D and Vep07. The molecular typing and cluster analysis were carried out by multi-locus molecular typing method. Results Of the 50 Vibrio vulnificus, 43 strains (43/50, 86%) carried plasmids to form 19 kinds of plasmids: 20 strains (20/50, 40%) had virulence and binding plasmids simultaneously, 13 strains / 50, 26%) only virulence plasmid, 2 (2 / 50,4%) only binding plasmid exists. Multi-site sequence typing method will be 50 strains of Vibrio vulnificus, divided into 43 ST type. Conclusion Plasmids are widely found in Vibrio vulnificus isolated from environmental samples in Nanshan District, Shenzhen. Plasmids carried by different types of isolates are different and have genetic diversity.
