目的观察吡格列酮对胰岛素抵抗SD大鼠胰腺组织中蛋白酪氨酸磷酸酶1B(PTP-1B)表达的影响。方法选取SD大鼠60只,随机分为对照组(NC组,n=20),喂养普通饲料;高脂组(HF组,n=40)同时分为高脂对照组(HFN组,n=20)和高脂吡格列酮组(HFP组,n=20),喂养高脂饲料12周后,进行模型验证。HFP组给予吡格列酮30 mg/(kg.d)灌胃2周,NC组和HFN组给予相同体积的生理盐水灌胃,应用免疫组化和western blot技术测定大鼠胰腺组织中PTP-1B蛋白的表达。结果 HF组大鼠胰岛素敏感性较NC组大鼠明显降低,差异有统计学意义(P<0.01);HFN组、HFP组胰腺组织PTP-1B表达量较NC组明显增高(P<0.01),与HFN组相比,HFP组PTP-1B表达量明显减少(P<0.01)。结论高脂饮食诱导的肥胖大鼠产生胰岛素抵抗,同时使其胰腺中PTP-1B蛋白表达量升高。吡格列酮可以降低胰腺组织中PTP-1B的表达,改善胰岛素抵抗。
Objective To observe the effect of pioglitazone on the expression of protein tyrosine phosphatase 1B (PTP-1B) in pancreatic tissue of insulin resistance SD rats. Methods Sixty Sprague Dawley rats were randomly divided into control group (n = 20) and NC group (n = 20). The rats in HF group (n = 40) 20) and high-fat pioglitazone group (HFP group, n = 20). After fed with high-fat diet for 12 weeks, model validation was carried out. HFP group was treated with pioglitazone 30 mg / (kg.d) for 2 weeks, NC group and HFN group were given the same volume of saline, the expression of PTP-1B protein in rat pancreas was detected by immunohistochemistry and western blot expression. Results The insulin sensitivity of HF group was significantly lower than that of NC group (P <0.01). The expression of PTP-1B in HFN group and HFP group was significantly higher than that of NC group (P <0.01) Compared with HFN group, the expression of PTP-1B in HFP group decreased significantly (P <0.01). Conclusion Obese rats induced by high-fat diet produce insulin resistance and increase the expression of PTP-1B protein in the pancreas. Pioglitazone can reduce the expression of PTP-1B in pancreatic tissue and improve insulin resistance.
