目的 探讨鼠S1 80 细胞HSP 70多肽诱导杀伤性T淋巴细胞 (CTL)及其特异性抗瘤作用机制。方法 从 43℃ 9h热休克诱导HSP高表达的S1 80 细胞提纯 80 μg mlHSP 70多肽 ,体外刺激小鼠脾淋巴细胞扩增、活化 ,然后用MTT法测定其杀瘤活性 ,并用电镜观察其培养上清诱导S1 80 细胞的形态学变化。结果 S1 80 细胞HSP 70多肽体外免疫扩增的小鼠脾淋巴细胞对S1 80 和H2 2 细胞的杀瘤率分别为83 .61%和 9.98% ,其培养上清能诱导S1 80 细胞出现程序化死亡的形态学变化。结论 S1 80 细胞HSP 70多肽能剌激小鼠脾淋巴细胞产生特异性杀伤S1 80 细胞的免疫活性细胞 ,该细胞还能分泌免疫活性物质诱导肿瘤细胞凋亡。
Objective To investigate the mechanism of HSP70 polypeptide inducing killer T lymphocyte (CTL) and its specific antitumor activity in murine S180 cells. Methods 80 μg ml HSP70 peptide was purified from heat-shock-induced HSP80 cells at 43 ℃ for 9 h, stimulated splenic lymphocyte proliferation and activation in vitro, and then its cytotoxicity was measured by MTT assay. Qing induced S180 cell morphological changes. Results The killing rates of S180 and H2 2 cells stimulated by HSP70 polypeptide in vitro were 83.61% and 9.98%, respectively. The culture supernatant of S180 cells induced the programmed cell apoptosis Morphological changes in death. Conclusion The HSP70 polypeptide of S180 cells can stimulate the splenic lymphocytes of mice to produce immunocompetent cells that specifically kill S180 cells. The cells can also secrete immunocompetent substances to induce tumor cell apoptosis.
