目的:评价生长分化因子11(growth differentiation factor 11,GDF11)对小鼠骨髓间充质干细胞(bonemarrow mesenchymal stem cells,BMSCs)骨向分化的影响。方法:BMSCs随机分为对照组和实验组,成骨诱导培养后,行碱性磷酸酶(ALP)活性检测和Von Kossa染色,实时荧光定量逆转录PCR(qRT-PCR)检测成骨相关基因,Western blot检测Smad的磷酸化水平。结果:GDF11降低ALP活性,减少胞外基质矿化,并通过激活Smad信号通路下调成骨相关基因的表达。结论:GDF11可抑制BMSCs骨向分化。
Objective: To evaluate the effect of growth differentiation factor 11 (GDF11) on bone differentiation of mouse bone marrow mesenchymal stem cells (BMSCs). Methods: BMSCs were randomly divided into control group and experimental group. After osteogenic induction, alkaline phosphatase (ALP) activity assay and Von Kossa staining were performed. Real-time quantitative reverse transcription PCR (qRT-PCR) Western blot detection of Smad phosphorylation level. Results: GDF11 decreased ALP activity, decreased extracellular matrix mineralization, and down-regulated the expression of osteogenesis-related genes by activating Smad signaling pathway. Conclusion: GDF11 can inhibit the bone differentiation of BMSCs.
