目的:观察有自发性巨核细胞集落(CFU-MK)形成的骨髓增殖性疾病(MPD)患者,其液体培养是否也有自发性巨核细胞生长。方法:采用末梢血非粘附单个核细胞进行体外无血清液体培养及半固体培养,应用RT-PCR技术检测液体培养自发性生长细胞的c-mpl及GPⅡb的表达。结果:只有在半固体培养基上形成自发性巨核细胞集落的患者,其无血清液体培养才有自发性生长的细胞,且两者为正相关(r=0.79, P<0.01),同时这些自发性生长的细胞有巨核细胞所特异的c-mpl和GPⅡb mRNA的平行表达。结论:无血清液体培养自发性生长的细胞中有巨核细胞的存在,与自发性CFU-MK来源于相同的巨核祖细胞,提示可利用前者代替集落细胞研究MPD自发性巨核细胞造血的机制。“,”Objective: Our purpose was to investigate if there are spontaneous megakaryocyte growth in liquid culture in vitro in patients with myeloproliferative disorders (MPD) who had spontaneous megakaryocyte colony (CFU-MK) formation. Methods: Non-adherent mononuclear cells (NMNC) from peripheral blood of the patients were cultured in serum-free medium and the expressions of c-mpl and GP-Ⅱb were examined by means of RT-PCR technique in the spontaneous growth cells. Results: There were spontaneous growth cells in serum-free liquid culture only in the patients who had spontaneous megakaryocyte colonies. A positive relation existed between them (r=0.79, P<0.01). There were parallel expressions of c-mpl and GPⅡb, which is specific to megakaryocytes, in the spontaneous growth cells. Conclusion: Megakaryocytes existed in the spontaneous growth cells in serum-free liquid culture, and derived from the same megakaryocyte progenitors as spontaneous CFU-MK, which indicated that the former would displace the spontaneous colony cells in the study on the mechanisms of spontaneous megakaryocytopoiesis in MPD.
