本研究对抗稻白叶枯病基因Xa23的标记03STS1进行改良和优化,设计分子标记命名为M-Xa23。以72个水稻亲本以及含抗性基因Xa23的CBB23与感病恢复系金刚30杂交的F2代分离群体为材料,并用白叶枯病Ⅶ型菌接种,结合抗病表型和标记结果,分析了M-Xa23的特异性和标记选择的准确性。实验结果表明:M-Xa23在抗病品种CBB23中扩增出200bp的特性条带,在其它71个水稻亲本中扩增出346bp的条带。144株F2分离群体中,109株表现抗病,35株表现感病,与标记M-Xa23检测的F2群体的基因型完全吻合,结果证明标记M-Xa23特异性强,能准确用于抗病基因Xa23的辅助选择。
In this study, the marker 03STS1 against rice bacterial leaf blight gene Xa23 was modified and optimized. The designed molecular marker was named M-Xa23. F2 segregating populations of 72 rice parents and CBB23 containing resistance gene Xa23 and susceptible restorer line Jingang30 were used as materials and inoculated with the type Ⅶ of Bacterial blight and combined with the resistance phenotype and the marker results M-Xa23 specificity and accuracy of marker selection. The results showed that M-Xa23 amplified a 200bp band in CBB23 and a 346bp band in 71 other rice cultivars. Of the 144 F2 isolates, 109 were resistant to disease and 35 were susceptible to disease. They were completely consistent with the F2 population detected by M-Xa23. The results showed that the M-Xa23 marker was highly specific and could be used accurately in disease resistance Auxiliary selection of gene Xa23.
