目的 从基因表达水平研究高山红景天苷对大鼠心脏内分泌功能的影响。方法 分别给各组大鼠按 15mg·kg- 1 ·d- 1 ip红景天苷氯化钠溶液。饲养 30d后 ,断头处死。冷酚法提取心房总RNA ,用 (α 32 P)标记的大鼠心钠素前体原cDNA控针与各组大鼠心房总RNA做Northernblot杂交和Dotblot杂交 ,以检测心房ANP mRNA的含量。结果 老年大鼠ANP mRNA含量明显低于青年大鼠 ,注射红景天苷后 ,老年大鼠ANP mRNA含量增加而青年大鼠减少。结论 老化过程中ANP基因表达能力明显衰退 ,红景天具有一定的抑制心脏内分泌功能老化的作用。
Objective To study the effect of salidroside on the endocrine function of rat heart from the level of gene expression. Methods Rats were given 15 mg·kg - 1 ·d - 1 ip salidroside sodium chloride solution. After 30 days of feeding, they were killed in a broken head. The atrial total RNA was extracted by cold phenol method. Northern astroblast hybridization and dotblot hybridization were performed with a (32P)-labeled rat atrial natriuretic precursor cDNA control needle and atrial total RNA from each group to detect the content of atrial ANP mRNA. Results The ANP mRNA content of aged rats was significantly lower than that of young rats. After injecting salidroside, the ANP mRNA content of aged rats increased and that of young rats decreased. Conclusion The ability of ANP gene expression declines significantly during aging, and Rhodiola has a certain effect of inhibiting the aging of cardiac endocrine function.
