目的研究胞嘧啶脱氨酶基因 (CD)联合化疗药物对结肠癌的治疗作用。方法以重组腺病毒AdCMVCD为载体转移CD基因 ,在体外研究病毒感染对顺铂 (CDDP)、丝裂霉素 (MMC)敏感性的影响 ,化疗药物对AdCMVCD/ 5 FC直接杀伤作用及“旁观者效应”的影响。结果病毒感染并不影响Lovo细胞对CDDP、MMC的敏感性 ;低剂量CDDP、MMC即可明显增强AdCMVCD/ 5 FC系统的直接杀瘤作用 ,5 FC的IC50 明显降低 (P <0 0 5 ) ,并且显著增强AdCMVCD / 5 FC系统的“旁观者效应”。结论化疗药物可以增强自杀基因系统对结肠癌的治疗作用 ,自杀基因和化疗药物有望联合应用于结肠癌的治疗
Objective To study the therapeutic effect of cytosine deaminase gene (CD) combined with chemotherapy on colon cancer. Methods The recombinant adenovirus AdCMVCD was used as a vector to transfer CD gene. The effects of virus infection on the sensitivity of cisplatin (CDDP) and mitomycin C (MMC) were investigated in vitro. The direct killing effect of chemotherapeutic drugs on AdCMVCD / 5 FC and “bystander Effect ”of the impact. Results The virus infection did not affect the sensitivity of Lovo cells to CDDP and MMC. The direct cytotoxicity of AdCMVCD / 5 FC system was enhanced by low dose of CDDP and MMC, and the IC50 of 5 FC was significantly lower (P <0.05) And significantly enhance the “bystander effect” of the AdCMVCD / 5 FC system. Conclusion Chemotherapy can enhance the therapeutic effect of suicide gene system on colon cancer. Suicide gene and chemotherapeutic drugs are expected to be applied to the treatment of colon cancer
