目的 探讨听神经瘤中神经生长因子受体的表达与肿瘤自然生长率、细胞增殖和血管形成的关系。方法 运用免疫组化技术,检测34 例人听神经瘤中神经生长因子受体蛋白、增殖细胞核抗原和微血管密度的表达。分析神经生长因子受体与增殖细胞核抗原、微血管密度及听神经瘤生长率的关系。结果 神经生长因子受体在所有肿瘤组织中表达(100 % ) ,而在正常听神经组织中无表达。肿瘤明显生长组神经生长因子受体、增殖细胞核抗原表达水平和微血管密度平均值显著高于缓慢或无生长组( P< 005 , P< 005 , P< 001) 。神经生长因子受体且和微血管密度( P< 001) 密切相关,但与增殖细胞核抗原( P> 005) 无关。结论 神经生长因子受体在明显生长的听神经瘤中表达增强,且与肿瘤的血管生成密切相关,提示神经生长因子受体参与了听神经瘤的发生过程。
Objective To investigate the relationship between the expression of nerve growth factor receptor and the natural tumor growth rate, cell proliferation and angiogenesis in acoustic neuroma. Methods Immunohistochemistry was used to detect the expression of nerve growth factor receptor protein, proliferating cell nuclear antigen and microvessel density in 34 cases of human acoustic neuroma. The relationship between nerve growth factor receptor and proliferating cell nuclear antigen, microvessel density and acoustic neuroma growth rate was analyzed. Results NGFR was expressed in all tumor tissues (100%) but not in normal auditory nerve tissue. The average expression of nerve growth factor receptor, proliferating cell nuclear antigen expression and microvessel density in the tumor-growth group was significantly higher than that in the slow or no-growth group (P <005, P <005, P <001). Nerve growth factor receptor and microvessel density (P <0 01) are closely related, but not with PCNA (P> 0 05). Conclusion The expression of nerve growth factor receptor in obviously growing acoustic neuroma is closely related to tumor angiogenesis, suggesting that nerve growth factor receptor is involved in the process of acoustic neuroma.
