目的:探讨清燥救肺汤对荷CT26小鼠结肠癌增殖及侵袭转移相关蛋白核转录因子-κB(nuclear transcription factor kappa B,NF-κB),血管内皮生长因子(vascular endothelial growth factor,VEGF),血管内皮细胞生长因子受体-1(vascular endothelial growth factor receptor-1,VEGFR-1),基质金属蛋白酶-9(matrix metalloprotein-9,MMP-9)表达的影响。方法:将50只雄性BALB/c小鼠,随机分为模型组,化疗[50 mg·kg~(-1)·(2 d)~(-1)]组,清燥救肺汤高、中、低剂量(15.2,7.6,3.8 g·kg~(-1)·d~(-1))组,每组10只。小鼠右腋下注射CT26细胞建立结肠癌小鼠模型,清燥救肺汤组以相应剂量造模前2周开始灌胃给药,造模后化疗组以5-氟尿嘧啶[5-FU,50 mg·kg~(-1)·(2 d)~(-1)]腹腔注射给药,模型组以等体积生理盐水灌胃给药,造模后2周后处死各组小鼠并取瘤,称重计算抑瘤率,蛋白免疫印迹法(Western blot)检测NF-κB,VEGF,VEGFR-1及MMP-9蛋白表达。结果:与模型组比较,清燥救肺汤高、中剂量组瘤重显著减小(P<0.01)。化疗组及清燥救肺汤高、中、低剂量组抑瘤率分别为83.90%,60.98%,44.39%,21.46%。与模型组比较,清燥救肺汤高、中剂量组NF-κB及VEGF蛋白表达明显降低(P<0.05,P<0.01)。与模型组比较,清燥救肺汤高、中、低剂量组VEGFR-1及MMP-9蛋白表达明显降低(P<0.05,P<0.01)。结论:清燥救肺汤可能通过降低NF-κB,VEGF,VEGFR-1,MMP-9蛋白表达,发挥抑制荷CT26小鼠结肠癌细胞增殖及侵袭转移的功效。
Objective: To investigate the effect of Qingzao Yifei Decoction on the proliferation of colon cancer cells and the expression of nuclear transcription factor-kappa B (NF-κB), vascular endothelial growth factor (VEGF) , Vascular endothelial growth factor receptor-1 (VEGFR-1) and matrix metalloprotein-9 (MMP-9) Methods: 50 male BALB / c mice were randomly divided into model group and chemotherapy group [50 mg · kg -1 (2 d) -1] , Low dose (15.2, 7.6 and 3.8 g · kg -1 · d -1) groups, 10 rats in each group. Mice were injected with CT26 cells in the right armpit to establish a mouse model of colon cancer. The rats in Zao Qing Yang Fei Decoction group were given gavage two weeks before the corresponding dosage. The model group was treated with 5-fluorouracil [5-FU, 50 mg · kg ~ (-1) · (2d) ~ (-1)] were intraperitoneally injected. The model group was given intragastric administration of an equal volume of normal saline, and the mice in each group were sacrificed 2 weeks after the model was established, , The tumor inhibition rate was calculated by weighing, and the protein expressions of NF-κB, VEGF, VEGFR-1 and MMP-9 were detected by Western blot. Results: Compared with the model group, the weight of the rats in high and medium dose Qingzaojiu Decoction group decreased significantly (P <0.01). The inhibition rates of chemotherapy, Qingzaojianfei Decoction high, medium and low dose groups were 83.90%, 60.98%, 44.39%, 21.46% respectively. Compared with the model group, the expressions of NF-κB and VEGF protein in Qingpi Yifei Decoction group were significantly decreased (P <0.05, P <0.01). Compared with the model group, the expressions of VEGFR-1 and MMP-9 in high, medium and low dose Qingzaojiu decoction group were significantly decreased (P <0.05, P <0.01). Conclusion: Qingzao Yifei Decoction may inhibit the proliferation, invasion and metastasis of colon cancer cells in CT26 mice by decreasing the expression of NF-κB, VEGF, VEGFR-1 and MMP-9.
