目的通过分析Semaphorin 3A(Sema3A)在类风湿关节炎(rheumatoid arthritis,RA)患者外周血(peripheral blood mononuclear cell,PBMC)及血清中的表达水平,探讨Sema3A在RA发病机制中的作用。方法 (1)收集RA患者130例,其它风湿病患者165例和健康对照者150例,采用酶联免疫吸附试验(enzyme-linked immuno sorbent assay,ELISA)检测血清中Sema3A的水平,并分析其与RA临床及实验室指标的相关性。(2)收集45例RA患者和35例健康对照的外周血PBMC,采用实时定量聚合酶链反应(polymerase chain reaction,PCR)法检测Sema3A mRNA的表达水平。结果 RA患者血清Sema3A水平为(15.89±8.58)ng/ml,显著高于健康对照组(6.96±2.62)ng/ml,(P<0.01)和其它风湿病患者组(4.35±3.57)ng/ml,(P<0.01);RA患者外周血PBMC Sema3A mRNA水平为(1.83±1.47),较健康对照组(1.01±0.85)显著升高(P<0.01。血清Sema3A水平与血小板计数(r=0.229,P<0.01)、ESR(r=0.174,P<0.05)、IgM(r=0.254,P<0.01)、类风湿因子(RF)(r=0.299,P<0.01)、HRF-IgG(r=0.203,P<0.05)呈正相关。另外,RF阳性、HRF-IgG阳性、APF阳性、AKA阳性或抗CCP抗体阳性RA患者血清Sema3A水平均显著高于相应抗体阴性的患者(P<0.05。结论 Sema3A在RA患者血清及外周血PBMC中均表达增高,并与RA病情活动密切相关,提示Sema3A可能成为RA治疗潜在的新靶点。
Objective To investigate the role of Sema3A in the pathogenesis of RA by analyzing the expression of Semaphorin 3A (Sema3A) in peripheral blood mononuclear cells (PBMC) and serum in patients with rheumatoid arthritis (RA). Methods (1) 130 cases of RA, 165 cases of other rheumatic diseases and 150 cases of healthy controls were collected. Serum levels of Sema3A were detected by enzyme-linked immunosorbent assay (ELISA) RA clinical and laboratory indicators of relevance. (2) Peripheral blood PBMCs were collected from 45 patients with RA and 35 healthy controls. The expression of Sema3A mRNA was detected by real-time polymerase chain reaction (PCR). Results Serum level of Sema3A in RA patients was significantly higher than that in healthy controls (6.96 ± 2.62 ng / ml, P <0.01) and other rheumatic diseases patients (4.35 ± 3.57) ng / ml (P <0.01). The level of Sema3A mRNA in peripheral blood of RA patients was significantly higher than that of healthy controls (1.83 ± 1.47 vs 1.01 ± 0.85, P <0.01). The levels of Sema3A and platelet count (r = 0.229, (R = 0.299, P <0.01), HRF-IgG (r = 0.203, P 0. 01), ESR (r 0.174, , P <0.05) .In addition, the levels of serum Sema3A in RF positive, HRF-IgG positive, APF positive, AKA positive or anti-CCP positive RA patients were significantly higher than those in corresponding antibody negative patients (P <0.05) .Conclusion Sema3A The expression of both the serum and peripheral blood PBMC of patients with RA was increased, which was closely related to the activity of RA, suggesting that Sema3A may be a potential new target for RA therapy.
