目的研究补阳还五汤、少腹逐瘀汤和丹参饮对气虚血瘀型大鼠红细胞膜蛋白组成的影响。方法 80只Wistar大鼠,随机分为8组,分别为空白对照组、气虚血瘀模型组、补阳还五汤高低剂量组、少腹逐瘀汤高低剂量组、丹参饮高低剂量组,每组10只。大鼠采取饥饿加力竭游泳法复制气虚血瘀模型,每日1次,连续14天。末次造模后禁食不禁水24 h。每日1次灌胃给药,连续给药15天。取血制备红细胞膜,采用SDS-PAGE法分析红细胞膜蛋白组成变化。结果与空白组比较,模型组带5及带6蛋白含量显著降低(P<0.05)。与模型组比较,补阳还五汤可显著升高带5蛋白(P<0.05)、带6蛋白(P<0.05)和带7蛋白含量(P<0.05);少腹逐瘀汤可显著升高带2蛋白(P<0.05)、带3蛋白(P<0.05)、带5蛋白(P<0.01)和带6蛋白含量(P<0.05);丹参饮可显著升高带4.1蛋白(P<0.05)、带5蛋白(P<0.05)、带6蛋白(P<0.05)和带7蛋白含量(P<0.05)。结论补阳还五汤、少腹逐瘀汤和丹参饮可以不同程度地纠正气虚血瘀模型大鼠红细胞膜的膜蛋白组成。
Objective To study the effects of Buyang Huanwu Decoction, Shaofuzhuyu Decoction and Salvia Decoction on erythrocyte membrane protein composition in rats with Qi-deficiency and Blood-stasis syndrome. Methods Eighty Wistar rats were randomly divided into eight groups: blank control group, Qi-deficiency and blood-stasis model group, Buyang Huanwu decoction high-low dose group, Shaofuzhuyu decoction high-dose group and Danshenyin high-dose group Group of 10. Rats take hunger and exhausting swimming copy qi deficiency and blood stasis model, 1 day, for 14 days. After the last modeling fast can not help but water 24 h. Gavage once daily administration, continuous administration of 15 days. Blood was collected to prepare erythrocyte membranes, and the protein composition of erythrocyte membrane was analyzed by SDS-PAGE. Results Compared with the blank group, the content of 5 and 6 in the model group was significantly decreased (P <0.05). Compared with the model group, Buyang Huanwu Decoction could significantly increase the expression of 5 protein (P <0.05), 6 protein (P <0.05) and 7 protein (P <0.05) (P <0.05), with 3 protein (P <0.05), with 5 protein (P <0.01) and with 6 protein (P <0.05) 0.05), 5 protein (P <0.05), 6 protein (P <0.05) and 7 protein (P <0.05). Conclusion Buyang Huanwu Decoction, Shaofuzhuyu Decoction and Danshen Decoction can correct the membrane protein composition of erythrocyte membrane of rats with Qi deficiency and blood stasis to varying degrees.
