目的:研究氯化两面针碱(nitidine chloride,NC)对人卵巢癌紫杉醇耐药细胞株A2780 Taxol细胞耐药性的逆转作用及机制。方法:氯化两面针碱干预A2780 Taxol后,应用MTT比色法检测多柔比星(adriamycin,ADM),依托泊苷(etoposide,VP-16),羟基喜树碱(hydroxycamptothecin,HCPT)对A2780Taxol细胞的抑制率;实时荧光定量PCR检测不同给药组对多药耐药基因(MDR1 mRNA)表达的影响;HE染色法观察肿瘤细胞结构的改变;TUNEL法分析其对细胞凋亡的影响。结果:耐药细胞A2780 Taxol对ADM,VP-16,HCPT的耐药倍数分别为1.87,1.07,3.25;经氯化两面针碱干预后,降低了A2780Taxol细胞对抗肿瘤药ADM,VP-16,HCPT的IC50值,可下降A2780 Taxol细胞的多药耐药(MDR1)基因的表达,HE染色可见NC给药组改变了A2780 Taxol细胞的形态结构,TUNEL染色凋亡细胞明显增多,凋亡指数为(58.03±1.46)%。结论:氯化两面针碱能有效逆转A2780 Taxol多药耐药性,具有良好的抗肿瘤药物多药耐药逆转作用。
Objective: To investigate the reversal effect and mechanism of nitidine chloride (NC) on the drug resistance of human ovarian cancer cell line A2780 Taxol. Methods: MTT assay was used to detect the effect of adriamycin (ADM), etoposide (VP-16) and hydroxycamptothecin (HCPT) on A2780Taxol Cells were detected by real-time fluorescent quantitative PCR. The expression of multidrug resistance gene (MDR1 mRNA) in different groups was detected by real-time fluorescent quantitative PCR. The changes of tumor cell structure were observed by HE staining and the cell apoptosis was analyzed by TUNEL. Results: The resistant rates of A2780 Taxol to ADM, VP-16 and HCPT were 1.87, 1.07 and 3.25, respectively. The A2780Taxol cells treated with A2780Taxol inhibited the proliferation of ADM, VP-16 and HCPT IC50 value of A2780 Taxol cells could decrease the expression of multidrug resistance (MDR1) gene in A2780 Taxol cells. HE staining showed that the morphology of A2780 Taxol cells was changed by HE staining. The apoptotic index of TUNEL-stained cells was significantly increased 58.03 ± 1.46)%. CONCLUSION: Nitidine chloride can effectively reverse A2780 Taxol multidrug resistance and has a good reversal effect of multidrug resistance of antitumor drug.
